RESUMO

Melanoma is the most aggressive and prevalent form of skin cancer globally, with a higher incidence in men and individuals with fair skin. Early detection of melanoma is essential for the successful treatment and prevention of metastasis. In this context, deep learning methods, distinguished by their ability to perform automated and detailed analysis, extracting melanoma-specific features, have emerged. These approaches excel in performing large-scale analysis, optimizing time, and providing accurate diagnoses, contributing to timely treatments compared to conventional diagnostic methods. The present study offers a methodology to assess the effectiveness of an AlexNet-based convolutional neural network (CNN) in identifying early-stage melanomas. The model is trained on a balanced dataset of 10,605 dermoscopic images, and on modified datasets where hair, a potential obstructive factor, was detected and removed allowing for an assessment of how hair removal affects the model's overall performance. To perform hair removal, we propose a morphological algorithm combined with different filtering techniques for comparison: Fourier, Wavelet, average blur, and low-pass filters. The model is evaluated through 10-fold cross-validation and the metrics of accuracy, recall, precision, and the F1 score. The results demonstrate that the proposed model performs the best for the dataset where we implemented both a Wavelet filter and hair removal algorithm. It has an accuracy of 91.30%, a recall of 87%, a precision of 95.19%, and an F1 score of 90.91%.

The organophosphate insecticide chlorpyrifos (CPF), an acetylcholinesterase inhibitor, has raised serious concerns about human safety. Apart from inducing synaptic acetylcholine accumulation, CPF could also act at nicotinic acetylcholine receptors, like the α7-isoform (α7-nAChR), which could potentially be harmful to developing brains. Our aims were to use molecular docking to assess the binding interactions between CPF and α7-nAChR through, to test the neurocytotoxic and oxidative effects of very low concentrations of CPF on SH-SY5Y cells, and to hypothesize about the potential mediation of α7-nAChR. Docking analysis showed a significant binding affinity of CPH for the E fragment of the α7-nAChR (ΔGibbs: -5.63 to -6.85 Kcal/mol). According to the MTT- and Trypan Blue-based viability assays, commercial CPF showed concentration- and time-dependent neurotoxic effects at a concentration range (2.5-20 µM), ten-folds lower than those reported to have crucial effects for sheer CPF. A rise of the production of radical oxygen species (ROS) was seen at even lower concentrations (1-2.5 µM) of CPF after 24h. Notably, our docking analysis supports the antagonistic actions of CPF on α7-nAChR that were recently published. In conclusion, while α7-nAChR is responsible for neuronal survival and neurodevelopmental processes, its activity may also mediate the neurotoxicity of CPF.

Acetamiprid (ACE) and Imidacloprid (IMI) are widely-used neonicotinoid insecticides (NNIs) with functional activity at human acetylcholine nicotinic receptors and, therefore, with putative toxic effects. The objective of this study was the evaluation of the interactions between NNIs and α7-nAChR, as this receptor keeps intracellular Ca2+ ([Ca2+]i) to an optimum for an adequate neuronal functioning. Possible interactions between NNIs and the cryo-EM structure of the human α-7 nAChR were identified by molecular docking. Additionally, NNI effects were analyzed in neuroblastoma SH-SY5Y cells, as they naturally express α-7 nAChRs. Functional studies included proliferative/cytotoxic effects (MTT test) in undifferentiated SH-SY-5Y cells and indirect measurements of [Ca2+]i transients in retinoic acid-differentiated SH-SY-5Y cells loaded with Fluo-4 AM. Docking analysis showed that the binding of IMI and ACE occurred at the same aromatic cage that the specific α-7 nAChR agonist EVP-6124. IMI showed a better docking strength than ACE. According to the MTT assays, low doses (10-50 µM) of IMI better than ACE stimulated neuroblastoma cell proliferation. At higher doses (250-500 µM), IMI also prevailed over ACE and dose-dependently triggered more abrupt fluorescence changes due to [Ca2+]i mobilization in differentiated SH-SY5Y neurons. Indeed, only IMI blunted nicotine-evoked intracellular fluorescence stimulation (i.e., nicotine cross-desensitization). Summarizing, IMI demonstrated a superior docking strength and more robust cellular responses compared to ACE, which were likely associated with a stronger activity at α-7nAChRs. Through the interaction with α-7nAChRs, IMI would demonstrate its high neurotoxic potential for humans. More research is needed for investigating the proliferative effects of IMI in neuroblastoma cells.

Neonicotinoids are effective insecticides with specificity for invertebrate nicotinic acetylcholine receptors. Neonicotinoids are chemically stable and tend to remain in the environment for long so concerns about their neurotoxicity in humans do nothing but increase. Herein, we evaluated the chronic toxic effects of acetamiprid- and imidacloprid-based insecticides over the differentiation of human neuroblastoma SH-SY5Y cells, which were exposed to these insecticides at a concentration range similar to that applied to crop fields (0.01-0.5 mM). Both insecticides did not have acute cytotoxic effects in both non-differentiated and in staurosporine-differentiated SH-SY5Y cells cytotoxicity as measured by the MTT and vital-dye exclusion tests. However, after a chronic (7-day) treatment, only imidacloprid dose-dependently decreased the viability of SH-SY5Y cells (F(4,39) = 43.05, P < 0.001), largely when administered-during cell differentiation (F(4,39) = 51.86, P < 0.001). A well-defined dose-response curve was constructed for imidacloprid on day 4 (R2 = 0.945, EC50 = 0.14 mM). During differentiation, either imidacloprid or acetamiprid dose-dependently caused neurite branch retraction on day 3, likely because of oxidative stress, to the extent that cells turned into spheres without neurites after 7-day treatment. Despite their apparent safety, the neurodevelopmental vulnerability of SH-SY5Y neurons to the chronic exposure to imidacloprid and to a lesser extent to acetamiprid points to a neurotoxic risk for humans.

Ascent to high altitude (> 3000 m height above sea level or m.a.s.l) exposes people to hypobaric atmospheric pressure and hypoxemia, which provokes mountain sickness and whose symptoms vary from the mild acute mountain sickness to the life-threatening, high-altitude pulmonary edema (HAPE). This study analysed the risk factors underlying HAPE in dwellers and travellers of the Ecuadorian Andes after sojourning over 3000 m height. A group of HAPE patients (N = 58) was compared to a NO HAPE group (N = 713), through demographic (ethnicity, sex, and age), red blood cell parameters (erythrocytes counts, hematocrit, median corpuscular volume, median corpuscular haemoglobin, and median corpuscular haemoglobin concentration (MCHC)), altitude (threshold: 3000 m.a.s.l.), and health status (vital signs) variables. Analysis of Deviance for Generalised Linear Model Fits (logit regression) revealed patterns of significant associations. High-altitude dwellers, particularly children and elder people, were HAPE-prone, while women were more tolerant of HAPE than men. Interestingly, HAPE prevalence was strongly related to an increment of MCH. The residence at middle altitude was inversely related to the odds of suffering HAPE. Ethnicity did not have a significant influence in HAPE susceptibility. Elevated MCHC emerges like a blood adaptation of Andean highlanders to high altitude and biomarker of HAPE risk.

Nitrogen (N) is a crop macronutrient of major importance, which affects both plant growth and yield. In this paper we discuss the humus content (%) and various soluble N forms (NO 3-, total N, nitrate-N, ammonium-N, and organic nitrogen) available in humus sand soil samples originating from the Pallag Experimental Station of Horticulture at the University of Debrecen, Hungary. We found 45.4% nitrate-N and 13.8% nitrite-N of total N content present in the soil. Considering the percentage distribution of soluble N forms present at the Pallag Experimental Station, we recommend using this soil in further pot experiments, given that this has optimal nutrient supply capacity. In addition, we examined possible statistical correlations between humus% and N forms.

Surface ozone is problematic to air pollution. It influences respiratory health. The air quality monitoring stations measure pollutants as surface ozone, but they are sometimes insufficient or do not have an adequate distribution for understanding the spatial distribution of pollutants in an urban area. In recent years, some projects have found a connection between remote sensing, air quality and health data. In this study, we apply an empirical land use regression (LUR) model to retrieve surface ozone in Quito. The model considers remote sensing data, air pollution measurements and meteorological variables. The objective is to use all available Landsat 8 images from 2014 and the air quality monitoring station data during the same dates of image acquisition. Nineteen input variables were considered, selecting by a stepwise regression and modelling with a partial least square (PLS) regression to avoid multicollinearity. The final surface ozone model includes ten independent variables and presents a coefficient of determination (R2) of 0.768. The model proposed help to understand the spatial concentration of surface ozone in Quito with a better spatial resolution.

The search for biotic compounds beyond planet Earth is among the highest priorities in space discovery. We sought to detect organic molecules in various earth soil samples collected from environments with extreme climatic conditions comparable to extraterrestrial environments using HPLC, spectrophotometry (UV, IR), GC/MS and vertical gel electrophoresis analyses systems. The applied methods in the study were compared using a self-developed software tool that was designed to evaluate the viability of instrument selection for organics detection in any given exploratory mission. The tool was created to aid further development and refinements of rapidly improving technological solutions such as MCE and MS instruments. Such comparison can be done quickly and easily, acting as a decision support tool when choosing between analytical methods during any planning phase of future exploration missions led by space agencies (i.e., NASA).

Soil management has great potential to affect soil respiration. In this study, we investigated the effects of organic versus conventional soil management on soil respiration.  We measured the main soil physical-chemical properties from conventional and organic managed soil in Ecuador. Soil respiration was determined using alkaline absorption according to Witkamp.  Soil properties such as organic matter, nitrogen, and humidity, were comparable between conventional and organic soils in the present study, and in a further analysis there was no statically significant correlation with soil respiration. Therefore, even though organic farmers tend to apply more organic material to their fields, but this did not result in a significantly higher CO2 production in their soils in the present study.

Strains of antibiotic-resistant bacteria have become more and more prevalent. This has attracted the attention of health agencies worldwide, leading to an urgent search for mechanisms to put a stop to this phenomenon. This study focuses on estimating the probability of a person in Ecuador (at potential risk) contracting an infection due to ampicillin-resistant Escherichia coli through the consumption of contaminated water, for which a residence area of people was considered in urban or rural areas. The analysis was carried out using the Bayes Theorem and the results show that in the rural population the probability of contracting an infection of this kind is 8.41% whilst in the urban area the probability is 3.57%. These results show an urgent need to provide safe water sources to the population, as well as to instigate an environmental legislation reform that allows for controlling the release of emerging pollutants, including antibiotics.

Since magnetic iron oxide nanoparticles (IONP) as magnetite (Fe3 O4 NPs) have potential applications in life sciences, industrial fields and biomedical care, the risks for occupational, general population and patients rises correspondingly. Excessive IONP accumulation in central nervous system (CNS) cells can lead to a disruption of normal iron metabolism/homeostasis, which is a characteristic hallmark resembling that of several neurodegenerative disorders. Fe3 O4 NPs- versus Fe3 O4 bulk-induced toxic effects have been assessed in two human CNS cells namely astrocytes (D384) and neurons (SH-SY5Y) after short-term exposure (4-24-48 h) to 1-100 µg ml-1 , and long-term exposure to lower concentrations. Short-term Fe3 O4 NPs induced significant concentration- and time-dependent alterations of mitochondrial function in D384 (25-75% cell viability decrease): effects started at 25 µg ml-1 after 4 h, and 1 µg ml-1 after 48 h. SH-SY5Y were less susceptible: cytotoxicity occurred after 48  h only with 35-45% mortality (10-100 µg ml-1 ). Accordingly, a more marked intracellular iron accumulation was observed in astrocytes than neurons. Membrane integrity was unaltered in both CNS cell types. Lowering Fe3 O4 NP concentrations (0.05-10 µg ml-1 ) and prolonging the exposure time (up to 10 days), D384 toxicity was again observed (colony number decrease at ≥0.05 µg ml-1 , morphology alterations and colony size reduction at ≥0.5 µg ml-1 ). Effects on SH-SY5Y appeared at the highest concentration only. Fe3 O4 bulk was always remarkably toxic toward both cells. In summary, human cultured astrocytes were susceptible to both Fe3 O4 NP and bulk forms following short-term and extended exposure to low concentrations, while neurons were more resistant to NPs. Cellular iron overload may trigger adverse responses by releasing iron ions (particularly in astrocytes) thus compromising the normal functions of CNS. Copyright © 2016 John Wiley & Sons, Ltd.

Several studies suggest that Iron Oxide nanoparticles may arrive to central nervous system independently of the route of administration. Actually, evidences indicate that the presence iron oxide nanoparticles into nervous system are linked to several neurodegenerative diseases. In this regard, our goal was to assess in vitro PolyVinylPirrolidone coated Iron Oxide nanoparticles, diameter of 20 nm, neuro-toxicity and their mechanism of action, which was fixed over the human neuronal cell line SH-SY5Y. Inducted biological effects were evaluated after 4­48 hours at crescents doses 1­100 µg/mL using the following endpoints: (i) Membrane integrity: Nanoparticles have produced no effect over cellular membrane for every dose and time evaluated; (ii) Mitochondrial activity: Starting at 10 µg/mL with a decrease of cellular vitality of 35%, and a maximum decrease of 45% at highest dose (100 µg/mL); (iii) Cellular morphology: Cells have evidenced no alteration after 48 hours of exposure; (iv) Cellular uptake: Dose-time dependent accumulation has observed: blue spots have been found at 10 µg/mL and over. Concluding, mitochondria are apparently the target: considering that the toxic effect produced by PolyVinylPirrolidone coated Iron Oxide nanoparticles after 48 hours of exposure in a dose-time dependent manner was evident.
